<b>Abstract:</b></div>

Cells constantly interpret extracellular signals and elicit appropriate responses through signaling networks of interacting proteins. Even in genetically identical cells in a homogenous environment, such networks are highly heterogeneous, and this variability
underlies e.g. cell-to-cell differences in the response to drugs of cancer cells.</div>

Recent advances in Single-cell protein profiling provide detailed snapshots of these signaling networks. In this talk, we present ongoing work on two computational approaches to model the heterogeneity in signal transduction.</div>

First, we introduce a graph-based method to detect protein–protein correlations that vary smoothly across the single-cell “state-space”, revealing how cell states can influence the way information propagates through the signaling network.</div>

Second, we describe a method to enable reconstruction of temporal trajectories from destructive measurements obtained at different timepoints after e.g. drug treatment. Specifically, we model cell states using Mixture of Factor Analyzers and align them across
timepoints using optimal transport.</div>

Our overall aim with these methods is to uncover context-dependent signaling patterns and connect cell state to cell fate, offering new insights into mechanisms of anti-cancer drug sensitivity and resistance.</div>
